I've noticed that a site with which I've exchanged links has linked to mydomain.co.uk/index.html rather than mydomain.co.uk/

I'm wondering whether this will devalue the positive effects of the link from a Google perspective?

If so, would it be a good idea for me to use .htaccess to redirect mydomain.co.uk/index.html to mydomain.co.uk/

Any help greatly appreciated.

Sure, why not! It can't hurt.
